Hi is there a Constraints manager tool to maintain a hole at a particular point on each corner of a frame so that when resizing the frame the holes stay at the point as required

Are we talking a 2D vector design or a 2.5/3d component?
Assuming 2D vectors there is no tool to do it in the same way as constraints as the hole would be resized with the frame but there are many alignment and moving options available for vectors which would do it. It's something I do often by using the alignment tools followed by the move tool set to absolute.

How about moving the holes to a new layer and lock that layer?

The original poster's description is a bit ambiguous so I know why you suggested what you did.
What the OP wants (as I interpret it) is for a circle to be attached to the vertex of the rectangle. e.g. if you have 1-inch square anchored at 0,0 and a circle anchored at 1,0 (the lower right corner of the rectangle) then when you resize the rectangle to 2 inches by 1 inch---keeping the origin at 0,0---the circle would automatically move to 2,0.
If you need that sort of functionality then other software are for you.

I take constraints as to what it is in Solidworks.
There always seems to be a feature in Vectric that I am not aware of, but I have not seen anything parametric or anything like constraints.
